It is a 31-amino acid analogue of human GLP-1 with a C18 fatty diacid chain enabling albumin binding, which extends its half-life to approximately one week — allowing once-weekly subcutaneous dosing. Semaglutide activates GLP-1 receptors in the pancreas, brain, and gut, resulting in enhanced insulin secretion, suppressed glucagon, delayed gastric emptying, and centrally mediated appetite suppression.
