5-AMINO-1MQ (Nicotinamide N-methyltransferase inhibitor) blocks NNMT, the enzyme that diverts SAM (S-adenosylmethionine) methyl groups away from NAD+ biosynthesis. NNMT inhibition elevates intracellular NAD+, activating the SIRT1-AMPK axis for enhanced fatty acid oxidation, mitochondrial biogenesis, and adipocyte lipolysis — reducing fat mass while preserving lean tissue. SLU-PP-332 is a potent agonist of ERRα (estrogen-related receptor alpha) and ERRγ — nuclear transcription factors that regulate the same gene programs activated by endurance exercise: OXPHOS (oxidative phosphorylation), PGC-1α signaling, mitochondrial biogenesis, and slow-twitch muscle fiber development. Together they create a dual-pathway exercise mimetic effect operating at both the metabolic substrate level (NAD+/NNMT) and the transcriptional level (ERR agonism).
