Primary Description
Kisspeptin increases the release of GnRH via the phospholipase pathway. PT-141 binds to melanocortin receptors in the hypothalamus. Both converge in increasing libido and sexual drive signals.

Kisspeptin-10 binds KiSS1R/GPR54 receptors on GnRH neurons in the hypothalamic arcuate and anteroventral periventricular nuclei, triggering pulsatile GnRH release and downstream LH/FSH surges. This restores testicular testosterone production in men and ovarian steroidogenesis in women — addressing hypogonadism at its hypothalamic origin. PT-141 (Bremelanotide) is a melanocortin receptor agonist: it activates MC3R and MC4R in the hypothalamus and limbic system to directly enhance sexual motivation, arousal, and erectile response through central dopaminergic and serotonergic pathways — independently of gonadal hormone levels.
